Police from the state's northern drug squad seized 48 kilograms of cannabis during raids on homes in two Launceston suburbs this week.
The street value of the drugs is estimated to be up to $350,000.
An man aged 22 and a 32-year-old man from Rosevears have been charged with trafficking.
They have been summonsed to appear in the Magistrates Court in Launceston at a later date.
Inspector Scott Flude from Launceston CIB believes the cannabis originated in Tasmania.
It is a very large seizure, it's very significant. I've never seen 48 kilos of dried cannabis in one area such as it was so it is significant as far as a drug seizure for Tasmania Police, he said.
My expectation is that seizing 48 kilograms of cannabis, especially for northern Tasmania, is a lot of cannabis, I think we'll see a noticeable impact on the street in the next few months.
Investigations are continuing.
